Overview

Viral proteases are crucial for successful viral replication. We will elucidate the details of their proteolytic autoactivation and search for protease-specific inhibitors. Our focus will mainly be on proteases from flaviviruses (Zika, Dengue), coronaviruses (SARS-CoV-2), and retroviruses (HIV).The project involves designing mutated protein variants, cloning, protein expression, and purification, along with screening compound libraries. These steps will be followed by biophysical characterization and crystallization. Additionally, we will focus on developing and adapting cell-based phenotypic assays and designing light-activatable protease variants.This work seeks to contribute to opening new paths to potentially innovative therapies.
